ONPATTRO® is a prescription medicaine used to treat adults with polyneuropathy (nerve damage) caused by hereditarty transthyretin-mediated (hATTR) amyloidosis. It belongs to a class of drugs called RNA interference therapy (sometimes called a gene silencer). ONPATTRO® was designated an 'orphan medicine' (a medicine used in rare diseases.
The active substance in ONPATTRO®, pastisiran, is a small interfering RNA' (siRNA), a very short piece of synthetic genetic material of the cell responsible for production of defective transthyretin. This reduces production of defective transthyretin, therby reducing the formation of amyloids and relieving the symptoms of hATTR amyloidosis.

ONPATTRO® is administered as an intravenous infusion in form of an IV drip. You'll get an ONPATTRO® dose every 3 weeks. The recommended dose is 300 micrograms per kilogram of body weight once every 3 weeks.
Pre-treatment may include medicines that help lower chances of infusion-related reactions such as corcticosteroid medicines, paracetamnol, and medicines known as H1 and H2 blockers before receiving ONPATTRO®.
Your doctor will monitor you for any infusion-related reactions during and after the infusion. If you have any infusion-related reaction, your doctor or nurse may slow down or stop your infusion, and you may need to take other medicines.
The most common side effects of ONPATTRO® include:
Treatment with ONPATTRO® lowers the amount of vitamin A inyour blood. Your doctor will tell you to take a vitamin A supplement every day. You should not take more than the amount of vitamin A recommended by your doctor. Low vitamin A levels can affect vision. If you develop problems with your vision (e.g., night blindness) while taking ONPATTRO®, talk to your doctor.
